Abstract: Objective To measure the expression level of miR-335-5p in the synovial fluid of pa-tients with osteoarthritis (OA)and analyze the relationship between miR-335-5p and Kellgren-Lawrence classi-fication grade.Methods The expression levels of miR-335-5p in the synovial fluid of OA patients (n =20) and non-OA controls (n =20)were detected using quantitative real-time PCR (qRT-PCR).Kellgren-Lawrence classification of the knee joint was evaluated.The correlation between miR-335-5p level and Kellgren-Lawrence classification grade was assessed.Results Down-regulated miR-335-5p was observed in synovial fluid of OA patients (△Ct 9.78 ±2.50)compared with that in non-OA controls (△Ct 8.23 ±2.13)(P <0.05).No correlation was observed between miR-335-5p level and Kellgren-Lawrence classification grade (rs =-0.351, P =0.062).Conclusion The expression level of microRNA-335-5p is down-regulated in the synovial fluid of OA patients,suggesting that it is probably correlated with the incidence and progression of OA.
