Preliminary study of serum specific miRNA for diagnosis of malignant melanoma

Abstract: Objective: To identify specific miRNA expression in the serum of melanoma patients and to evaluate its biological significance using a variety of analytic methods. Methods: The expression levels of specific miRNA in the serum of melanoma patients were measured with the low-throughput chips and real-time quantitative reverse transcription-polymerase chain reaction(qRT-PCR). Their specificity and sensitivity were assessed using cluster analysis and risk score. The relationship between miRNA expression and patients’ survival rates was further analyzed. Results: Specific expressions of four miRNAs were detected in the serum of melanoma patients, with high sensitivity(83.3%)and specificity(85.0%). Among these miRNAs, the expression levels of miR-222 were negatively correlated with the survival rate of melanoma patients. Conclusion: Specific expressions of four miRNAs are identified in the serum of melanoma patients. These miRNAs could be the specific biomarkers of melanoma.
