Study on the Half-life of Testis Germline Stem Cells in Three Lines of Drosophila

Abstract: The number of germline stem cells(GSCs)in Drosophila testis decreases with aging.Using immuno-fluores-cent staining technique,the testis GSCs number of three lines of Oregon,W1118 and Stat mutant flies is studied in different stage(day 1,15,30,45,60 and day 75 after eclosion)at room temprature.The average GSCs number of the aboved flies in day 1 after eclosion is respectively 8.55,8.60 and 8.15,the half-life of the two wild-type lines(Oregon,W1118)and the Stat mutant line are respectively sixty days and thirty days based on the curve generated on the relationship of the GSCs number and the elapsed days.There is a big difference in the half-life of testis GSCs between the wildtype and Stat mutant flies.The result indicates that gene Stat mutation dramatically accelerate the aging of GSCs in Drosophila testis.
