The expression and significance of RUNX3 in actinic keratosis, Bowen disease and squamous cell carcinoma

Abstract: Objective:To investigate the expression of RUNX3 in actinic keratosis(AK), Bowen disease(BD) and squamous cell carcinoma(SCC), and to reveal the role of RUNX3 and its clinical significance in AK, BD and SCC. Methods: Immunohistochemical staining(SP method) was used to detect the expression of RUNX3 protein in normal skin(n=10), AK(n=30),BD(n=30) and SCC(well differentiated n=30, moderately differentiated n=30, and poorly differentiated n=30) groups. The average optical density of positive expression in each field of vision was quantitatively analyzed with Image-Pro Plus 6.0(IPP)image analysis software. Results: The expression levels of RUNX3 protein were significantly different among these tissues as determined by average optical density(0.1841 ±0.0022 for normal, 0.1922 ±0.0025 for AK, 0.1926 ±0.0019 for BD, 0.1943 ±0.0025 for well differentiated SCC, 0.1945±0.0028 for moderately differentiated SCC, and 0.1946±0.0033 for poorly differentiated SCC). Compared with normal controls, the expression levels of RUNX3 protein were significantly higher in AK, BD, and three SCC(well, moderately and poorly differentiated) groups(all P<0.05);The expression levels of RUNX3 protein were also significant higher in three SCC groups than in AK groups(P<0.05). The differences between BD and AK or among three SCC groups were no significance(P>0.05). Conclusion: Normal skin expresses negligible levels of RUNX3 protein. The expression levels of RUNX3 protein are higher in SCC than in BD and AK. RUNX3 may play an important role in the evolution of cutaneous SCC. RUNX3 may become one of important biological indicators in early diagnosis, differential diagnosis,malignant grading and prognostic evaluation of skin tumors.
