The mechanistic study on the influences of STAT3 and type Ⅰ interferon in the replication of varicella zoster virus

Abstract: Objective:To investigate the association of STAT3 and type I interferon(IFN) with the replication of varicella zoster virus(VZV). Methods: After infection of human embryonic lung fibroblasts(HELF) with VZV, virus plaque test was used to detect VZV titer in normal controls, STAT3 overexpression and STAT3 knockout group. Real time fluorescence quantitative polymerase chain reaction(PCR) was used to detect the gene expression of type I IFN(OAS、PKR、IRF3、STAT1) in three groups. Results: After infection of HELF with VZV, the expression levels of STAT3 increased significantly. Overexpression of STAT3 increased VZV infection(P<0.001), and decreased the expression of type Ⅰ IFN response genes(OAS, PKR,IRF3) and STAT1(P=0.000). In contrast, STAT3 knockout decreased VZV virus titers(P=0.000), and increased type Ⅰ IFN response genes(OAS, PKR, IRF3) and STAT1(P=0.000). Conclusions: In human embryonic lung fibroblast cells, overexpression of STAT3 gene can enhance VZV infection, and inhibit the anti-infection of type I IFN. The knockdown of STAT3 gene inhibits VZV infection, and increases the expression of type I IFN response genes.
