Abstract: 目的 体外研究姜黄素对丙烯酰胺(AA)所致人肝癌HepG2细胞细胞毒性和遗传毒性的保护作用.方法 AA处理组(细胞毒性实验设5,10,20,40和80mmol/L 5个剂量,微核试验设1.25和2.50 mmol/L 2个剂量),姜黄素保护组设0.63、1.25和2.50 μg/mL 3个剂量组,噻唑蓝法观察姜黄素对AA所致细胞毒性的保护作用;微核试验观察姜黄素对从所致遗传毒性的保护作用.结果 随着AA浓度的升高,HepG2细胞的存活率逐渐下降,与相应浓度AA处理组比较,2.50 μg/mL姜黄素保护组的细胞存活率升高,差异有统计学意义(P<0.05);1.25、2.50 mmol/L AA组HepG2细胞微核率分别为(41.3‰±5.0‰)和(46.0‰±4.0‰),明显高于溶剂对照组,差异有统计学意义(P<0.05).2.50 μg/ml姜黄素保护组细胞微核率明显低于1225和2.50 μg/ml AA组,差异有统计学意义(P<0.05).结论 姜黄素对AA所致HepG2细胞的细胞毒性和遗传毒性具有保护作用.
