The supernatant of melanoma cell culture inhibits interleukin-12 production in murine peritoneal macrophaes in vitro

Abstract: 目的:探讨M14黑素瘤细胞培养上清液(melanoma culture supematants,MCS)在体外对小鼠腹腔巨噬细胞分泌白介素(IL)-12的影响.方法:将小鼠腹腔巨噬细胞在体外分别以脂多糖(LPS)(10μg/mL)、MCS(100μg/mL)、PD98059(20μmol/L)等单独及共同作用,用酶联免疫吸附(ELISA)法检测培养上清液中巨噬细胞IL-12 p40的分泌,用免疫印迹(Westem blot)法分析巨噬细胞细胞外调节蛋白激酶(ERK)44/42/丝裂原激活蛋白酶(MAPK)、p38 MAPK和c-Jun氨基末端激酶/应激活化蛋白激酶(JNK/SAPK)蛋白表达.结果:MCS在体外能够显著抑制小鼠腹腔巨噬细胞产生IL-12 p40[(12.82±2.87)ng/mL],且培养上清液中IL-12水平的降低与细胞核磷酸化的ERK44/42蛋白表达增强相一致;此外,经ERK44/42特异性抑制剂PD98059预处理的巨噬细胞其IL-12 p40的分泌量却不受MCS抑制[(25.23±4.52)ng/mL,P>0.05].结论:黑素瘤细胞分泌物通过活化ERK MAPK信号传导通路抑制小鼠腹腔巨噬细胞分泌IL-12.
