Mechanism of hirudin in brain edema after intracerebral hemorrhage

Abstract: 目的探讨凝血酶在脑出血后脑水肿形成中的作用和水蛭素抗水肿的作用.方法健康Wistar大鼠80只,随机分为四组,各组分3、7 d两个亚组,每组20只.A组:生理盐水+生理盐水;B组:生理盐水+水蛭素;C组:血+生理盐水;D组:血+水蛭素.采用立体定向注射方法,用微量注射器在右侧尾状核部位,注入自体未凝血(C、D组)或等量生理盐水(A、B组)50 μl,留置20 min后同部位二次注射10 μl生理盐水(A、C组)或等量水蛭素溶液(B、D组,浓度2 U/μl).术后第3、7天断头取脑,测定脑水含量.结果大鼠脑出血后脑组织含水量随时间推移而升高、变化,而水蛭素可减轻其水肿程度(P<0.05).结论凝血酶可以引起脑出血后脑组织水肿,水蛭素可以抑制凝血酶所引起的脑水肿且水蛭素本身不对脑组织产生负面影响.
