The Effects of Parathyroid Hormone on Calcium Mobilization in Rat Cardiomyocytes

Abstract: 目的:研究甲状旁腺素(PTH)对心肌细胞钙离子移动的影响及其机制.方法:培养的新生大鼠心肌细胞以荧光指示剂Fluo-3/AM负载,通过激光共聚焦显微镜(LSCM)动态观察不同浓度PTH1~34对培养心肌细胞钙移动的作用,以及细胞外钙和钙拮抗剂的影响.结果:在细胞外Ca2+为2.50 mmol/L时,PTH1~34在0.00、0.01、0.10和1.00 μmol/L浓度下的刺激可使心肌细胞静息钙荧光强度(FI)分别从48.78±6.70、47.78±6.14、45.58±6.11、51.10±9.33,增高至48.82±6.31(P>0.05)、54.63±3.60(P<0.05)、63.82±9.21(P<0.01)、78.66±10.04(P<0.01);1.00μmol/L浓度PTH1~34刺激,在细胞外液无钙时心肌细胞静息钙FI仅从48.88±7.06上升到55.50±6.52(P>0.05);如应用10.00μmol/L硝苯地平预处理,则FI从47.42±8.80上升到54.12±5.13(P>0.05).结论:甲状旁腺素显著增加静息心肌细胞[Ca2+]i,并呈浓度依赖性,电压依赖性钙通道开放引起的细胞外钙内流为其重要机制之一.
