Abstract: 目的: 制备抗p185erbB2单克隆抗体,并研究其对表达p185erbB2肿瘤细胞的结合及生长抑制作用.为后续工作奠定基础.方法:用NIH3T3-erbB2细胞免疫BALB/c小鼠,制备抗p185erbB2单克隆抗体.经ELISA、免疫沉淀、Western blot及免疫组化检测单抗的结合特异性.并用MTT比色法检测所获抗体对表达p185erbB2肿瘤细胞的生长抑制作用.结果:经筛选获得1株对表达p185erbB2细胞反应阳性,不表达p185erbB2细胞反应阴性的单克隆抗体1H3;免疫沉淀、Western blot均表明1H3可与p185erbB2分子特异性结合;免疫组化结果显示1H3对c-erbB2阳性的乳癌组织呈特异性着染;MTT比色法检测表明1H3对不同的p185erbB2阳性肿瘤细胞均有不同程度的生长抑制作用.结论: 单克隆抗体1H3能与p185erbB2特异结合,并可不同程度地抑制NIH3T3-erbB2或SKBR3细胞的生长,除可应用于临床对p185erbB2的检测外,尚具有肿瘤生物治疗的潜在应用开发价值.
