Abstract: 目的研究利用CTLA4Ig阻断B7/CD28相结合,诱导同种异体小鼠移植心脏免疫耐受.方法建立同种异体小鼠移植心脏模型,研究组腹腔注射CTLA4Ig,观察小鼠移植的心脏存活天数和病理改变.结果研究组移植的心脏存活时间为(81.0±51.2)d,对照组为(7.8±1.3)d,两组差异有显著性(P<0.05).组织学改变:对照组可见大量淋巴细胞、单核细胞浸润,组织充血水肿,心肌变性、坏死,出血,病理分级为Ⅳ级.研究组术后7 d病理改变较轻,表现为局灶性血管周围和心肌间质炎症细胞浸润、局灶性心肌损伤,病理分级为Ⅰ~Ⅱ级.术后30 d 研究组可见部分心肌变性、水肿、纤维增生,炎细胞浸润,血管管壁增厚、水肿、炎细胞外渗,病理分级为Ⅱ~Ⅲ级.结论利用CTLA4Ig阻断B7/CD28相结合可诱导同种异体小鼠心脏移植免疫耐受.
