Abstract: Objective To establish the optimal extraction process of Danai Dysmenorrhea Paste. Methods The volatile oil was extracted from Cinnamomi cortex, Cyperi rhizoma and Artemisiae argyi folium by supercritical CO2 extraction. The optimal extraction process was screened by an orthogonal experiment with the contents of tanshinone Ⅱ A and salvianolic acid B in alcohol extract determined by HPLC as indexes. Results The optimal extraction conditions for Cinnamomi cortex were the extraction pressure of 35 MP, extraction temperature of 40 ℃ and CO2 flow rate of 30 kg/h. The optimal extraction conditions for Cyperi rhizoma were the extraction pressure of 30 MPa, extraction temperature of 45 ℃ and CO2 flow rate of 5.5 kg/h. The optimal extraction conditions for Artemisiae argyi folium were the extraction pressure of 20 MPa, extraction temperature of 30 ℃ and CO2 flow rate of 24 kg/h. The optimal conditions of ethanol extraction were reflux extraction for 3 times with 10-fold of 75 % ethanol, 1.5 h each time. Conclusion The optimal extraction process can be simple, repeatable and accurate, providing reference for the preparation of traditional Chinese medicine patch.
