Correlation between the INSR gene polymorphism and the chemosensitivity of epithelial ovarian cancer

Abstract: Objective To investigate the influence of insulin receptor (INSR) gene polymorphism in platinum-based chemotherapy response in epithelial ovarian cancer (EOC). Method A total of 339 patients who underwent post-operative chemotherapy were recruited. Tag single nucleotide polymorphism (tagSNP) of INSR gene was screened based on HapMap and available literature. Frequency distribution of the genotypes and alleles in INSR gene was se-quenced by ABI3100-Avant to compare the distribution of INSR gene polymorphism between chemosensitive group and chemoresistant group and to identify the genotype which might influence chemosensitivity in EOC patients. Result Compared with GC+CC genotype of rs2252673, GG of rs2252673 showed decreased platinum-based chemosensitivity in INSR gene;and compared with GG+GC genotype of rs3745546, CC of rs3745546 showed less chemosensitivity to platinum medications. Logistic analysis showed that the polymorphism of rs2252673 and rs3745546 were the main fac-tors influencing the chemosensitivity of EOC (P<0.05). Conclusion The polymorphisms of rs2252673 and rs3745546 of INSR gene may be associated with platinum-based chemosensitivity of EOC patients.
