Exendin-4 Improves Aβ31-35 Induced Per2 Rhythm Gene/Protein Abnormal Expression in HT22 Nerve Cells of Mouse

Abstract: 目的 探讨Exendin-4对β淀粉样蛋白31-35(Aβ31-35)所致HT22细胞Per2节律基因/蛋白表达的影响及机制.方法 HT22细胞分为Aβ31-35处理组(0,2.5,5.0,10.0 μmol/L),Exendin-4单独处理组,Exendin-4预处理组,Exendin-4+Exendin(9-39)预处理组,Exendin-4+Exendin(9-39)处理组,Ex-endin(9-39)单独处理组,以完全培养基培养的细胞为对照组.光镜下观察HT22细胞形态,MTT法检测细胞存活率,实时荧光定量PCR检测Per2基因的表达,免疫荧光染色观察GLP-1R及PER2蛋白表达.结果 5,10 μmol/L Aβ31-35处理后,细胞存活率较对照组显著降低(P<0.05).Exendin-4预处理后,5μmol/L Aβ31-35引起的细胞形态结构的改变明显改善,细胞存活率显著提高(P<0.05).Exendin-4预处理后,Per2基因/蛋白表达在CT(circadian time) 16时较Aβ31-35处理显著提高(P<0.05);Exendin-4+ Exendin (9-39)处理后,GLP-1R表达在CT16时较Exendin-4处理显著降低(P<0.05).Exendin-4+Exendin(9-39)预处理后,CT16时PER2蛋白荧光强度较Exendin-4预处理显著降低(P<0.05).结论 Exendin-4可能通过上调GLP-1R,改善Aβ31-35所致HT22细胞Per2节律基因/蛋白异常表达.
