Abstract: 目的 探索模拟失重下空间诱变大肠杆菌(T1-13)感染巨噬细胞RAW264.7后差异表达的microRNAs (miRNAs)及其意义.方法 采用第二代测序技术检测模拟失重染菌组与对照组巨噬细胞miRNAs差异表达,通过生物信息学预测差异miRNAs的靶基因,并对靶基因进行GO功能富集分析、KEGG信号通路分析以及STRING蛋白互作分析.结果 模拟失重染菌组与对照组间筛选出33个差异miRNAs,下调23个,上调10个.这些miRNAs互补结合的靶基因参与了MAPK通路、WNT通路、轴突导向以及TGF-β等信号通路,其中关键下调的靶基因是MAPK9,MAPK14,MAP3 K7,MAP3 K14,PIK3CA,PIK3CB,PIK3CD,MYC,JUN,SMAD4;关键上调的靶基因是AKT3,MAPK8,MAPK9,MAPK10,MAP2K1,PIK3CD.结论 模拟失重下空间诱变大肠杆菌感染巨噬细胞后miRNAs表达出现显著差异,这些miRNAs可能通过调控MAPK,WNT以及TGF-β等通路参与失重条件下炎症反应的发生.
