Abstract: Thiopeptide antibiotics are a class of sulfur-rich and highly modified polypeptide compounds,exhibiting potent activities against bacterial pathogens.As an archetypal bi-cyclic thiopeptide,thiostrepton has received great attention from researchers at home and abroad.This review introduces the biological activities,the action mechanisms and the biosynthetic studies of thiostrepton.A biosynthetic pathway engineering concept based on mutation and modification of the active structural moieties of thiostrepton is proposed,which could generate more thiostrepton analogues with better activities.According to the characteristics of thiostrepton biosynthesis,three main engineering strategies are classified and the corresponding examples are discussed.This review will provide a new perspective to the studies on the biosynthetic mechanisms and structural engineering of the thiostrepton-related natural products.
