Abstract: 目的探讨吗氯贝胺(MCB)在聚维酮(PVPκ30)中分散作用的热力学,研制MCB-PVPκ30固体分散体.方法采用溶解度法测定MCB的溶解度及其热力学参数,差示扫描量热分析(DSC)和X-射线衍射分析鉴定MCB在该固体分散体(SD)中存在状态,并进行体外溶出度研究.结果 MCB-PVPκ30表观稳定常数(Kc)在实验范围内随温度升高而减小,反应热(ΔH0)为-13.22 kJ/mol.经DSC、X-射线衍射分析,SD中药物晶体消失(1∶3),经统计学分析所有SD的溶出速率显著大于原药的溶出速率.结论 MCB在PVPκ30中分散作用是放热过程,MCB-PVPκ30SD提高了MCB的溶出度,有助于MCB SD的制备及开发具有高生物利用度的新剂型.
