Quantitative Determination of Gardenoside,Genipin-1-O-β-D-gentian glycosides,Crocin-1 and Crocin-3 in Mongolian Medicine Compound Xieriga-4 by RP-HPLC

Abstract: To establish a method for quantitative determination of gardenoside,genipin-1-O-β-Dgentian glycosides,crocin-1,and crocin-3in Mongolian medicine compound xieriga-4by RP-HPLC at the same time.The RP-HPLC procedure was performed on a chromatographic column of Elite C18(460mm×250mm,5μm)column eluted with acetonitrile(A)-0.4% phosphoric acid(B)at a flow rate of 1mL/min,a temperature of 30℃,and a detection wavelength of 238 nm for gardenoside and genipin-1-O-β-D-gentian glycosides and 380 nm for crocin-1and crocin-3.The injection volume of samples was 20μL.This approach allowed gardenoside,genipin-1-O-β-D-gentian glycosides,crocin-1,and crocin-3effectively separated.The linearity,precision,stability,and reproducibility were good in the respective detection range.The average recovery of genipin-1-O-β-D-gentian glycosides,gardenoside,crocin-1,and crocin-3(n=6)was 100.16%(RSD =0.69%),99.19%(RSD =1.29%),100.59%(RSD =0.80%),and 99.06%(RSD =1.02%),respectively.The method validation showed that it was a simple,fast,and accurate method with a high repeatability for simultaneous quantification of gardenoside,genipin-1-O-β-D-gentian glycosides,crocin-1,and crocin-3in mongolian medicine xieriga-4,which can be used for the quality control of Mongolian medicine Xieriga-4.
