X-rays radiation enhances the stem-like capability of CD133~- nasopharyngeal carcinoma

Abstract: This study was performed to investigate the stem-like capability changes of CD133 nasopharyngeal carcinoma after X-ray radiation.Human nasopharyngeal carcinoma cells were sorted by magnetics-activated cell sorting(MACS),and then the purity of CD133~+ and CD133~- cells was detected by flow cytometer(FCM).And the stem-like characteristics were verified by invasion assay and tumor xenograft models in nude mice.Subsequently,the CD133~-cells were treated with 4 Gy X-ray radiation,and the invasion assay and tumor xenograft models were used to investigate the variation of tumorigenesis.Additionally,the expressions of CD133,SOX2 and OCT4 mRNA were detected by RT-PCR.Data showed that,the purity of CD133~+ and CD133~-cells were97.72%±0.82%and 1.63%±0.66%,respectively.And,compared with CD133~- cells,CD133~+ cells demonstrated stronger invasion and tumorigenesis.After treatment with 4 Gy X-ray radiation,the invasion and tumorigenesis of CD133~- cells were enhanced,and the expressions of CD133,SOX2 and OCT4 mRNA were up-regulated.In conclusion,X-ray radiation can enhance the stem-like capability of CD133~- nasopharyngeal carcinoma in vitro.
