Abstract: Objective: To observe the effect of Melastoma dodecandrum Lour Oral Liquid on peripheral blood platelets count,bleeding time,clotting time,megacaryocyte in marrow,interleukin-4 and interferon-γ in primary immune thrombocytopenia(ITP) mice model. Methods: Sixty BALB/C mice were randomly divided into Melastoma dodecandrum Lour Oral Liquid high,middle and low doses groups,prednisone acetate group,model group and normal control group. ITP mice models were made by intraperitoneally injecting guinea pig-anti mice platelet serum(APS) every other day. Eight days after injecting the APS,the mice were fed with medicine once a day. The peripheral blood platelets count,bleeding time,clotting time,megacaryocyte in marrow,interleukin-4 and interferon-γ were detected after 14 days. Results: 1The peripheral blood platelets count in Melastoma dodecandrum Lour Oral Liquid high,middle and low doses groups were significantly higher than that in model group(P < 0. 05). The bleeding time,clotting time,megacaryocyte in Melastoma dodecandrum Lour Oral Liquid high,middle and low doses groups were significantly lower than those in model group(P <0. 05). The bleeding time in Melastoma dodecandrum Lour Oral Liquid high dose group was lower than that in model group(P < 0. 05). The peripheral blood platelets count in Melastoma dodecandrum Lour Oral Liquid high and middle doses groups were higher than those in low dose group(P < 0. 05). 2The levels of IL-4,mainly IFN-γ and IFN-γ/IL-4 in model group were higher than those in normal control group(P < 0. 05). The levels of IL-4,IFN-γ and IFN-γ/IL-4 in Melastoma dodecandrum Lour Oral Liquid high dose group were lower than those in model group(P < 0. 05).But in Melastoma dodecandrum Lour Oral Liquid middle and low dose group,only IFN-γ and IFN-γ/IL-4 were lower than those in model group(P < 0. 05). The levels of IL-4,IFN-γ and IFN-γ/IL-4 among three doses groups had no significant change. Conclusion: Melastoma dodecandrum Lour Oral Liquid can increase peripheral blood platelets count and relieve symptoms of bleeding in ITP mice model. High dose can increase the therapeutic effect. Melastoma dodecandrum Lour Oral Liquid can regulate the immune function of ITP by decreasing the levels of IFN-γ and IFN-γ/IL-4and modulating the Th1/Th2 imbalance.
