Development of PCR to Detect Porcine circovirus Type2 and its clinical Application

Abstract: Porcine circovirus type 2(PCV2) is one of the most important pathogens in swine, a PCR method was established by using a set of primers derived from the published nucleotide sequence of the PCV2genes(Rep) in Gen Bank. In order to confirm its specificity, the PCR was implemented by using the template DNA, and the expected products of 700 bp were only detected in PCV2,however no specific band was detected in other virus such as PPV, CSFV, PRV. The lowest DNA consistence which could be detected by the PCR method was 1.80 ng/μL. In order to further investigate the prevalence of PCV2, 14 clinic samples were detected. The data indicated that PCV2 in 6 samples were positive. The experiments indicate the PCR method could be used in diagnosis and epidemiology investigating.
