Determination the Monosaccharide Components of Three Common Bacterial EPS with Reverse Phase - high - performance Liquid Chromatography

Abstract: To identify the complex components of the bacterial extra - cellular polymeric substances(EPS) and their effects of corrosion and surface modified on the mineral fine particles,three common bacteria from soil, air and human body were chosen to be tested strains. They were silicate bacteria,Staphylococcus aureus and Esch-erichia coli. RP - HPLC method was applied to rapid determinate the monosaccharide components of bacterial EPS qualitatively and quantitatively. The results showed 8 monosaccharides were separated well under the detecting condition. The monosaccharides of silicate bacterial EPS included mannose and glucuronic acid,while Staphylo-coccus aureusˊs EPS included ribose and glucose and Escherichia coli EPS included ribose and galactose. The dif-ferent bacteria consumed different monosaccharides in its growth process. The results also showed that the EPS monosaccharide components of each bacteria appeared apparent difference. In this research a method with RP -HPLC was established to determine the monosaccharide components of bacterial EPS.
