Abstract: Objective To observe the effectd of apelin -13 on mice with brain edema after intracerebral hemorrhage (ICH), and to explore the relationship between apelin -13 and the expression of aquaporin -4 (AQP4), matrix metal-loproteinase-9 (MMP9).Methods Healthy male CD-1 mice were randomly divided into three groups:a sham-op-erated group, a normal saline (NS) group and an apelin-13 group.A mice model of ICH was established by injection of collagenase IV to the striatum .Then, 5 min before ICH 0.3 g/g apelin-13 or the same volume of normal saline was in-tracerebroventricularly administered .The cerebral water content ( CWC) was measured by the dry -wet method, while the amounts of AQP4 and MMP-9 were determined by Western blotting .Results Compared with the sham -operated group, the CWC stated to significantly increase in Group NS 6 h after ICH and continued for 48 h, reaching a peak 24 h after ICH.Compared with Group NS, the CWC was remarkably decreased in Group apelin -13 24 h and 48 h after ICH. Meanwhile , the amounts of AQP4 and MMP9 were down regulated by apelin -13 in the ICH area and its surrounding tis-sues.Conclusion Apelin-13 can attenuate brain edema after ICH in mice , which may be associated with the down -regulation of AQP4 and MMP9 protein expression .
