People Hashimoto Thyroiditis miRNAs in Thyroid Organization Express the Differences of Spectral Analysis

Abstract: Objective: Hashimoto thyroiditis is a kind of autoimmune thyroid disease, and there is an upward trend in the incidence of this disease. In medical practice, HT is believed to be the most common cause of primary hypothyroidism and is considered to be related to development of both thyroid carcinoma and lymphoma. Because of the lack of early diagnostic criteria of HT, and its diversity and performance is too difficult to detect, the patient always suffer more delay treatment with poor outcomes. This paper aims to screen the thyroid tissue of HT lesions and reveal the changes of miRNA expressions of HT by microRNA microarray technology system.Methods: With miRNA array, we compared miRNA expressions of different normal thyroid tissue with HT. Results: The results of miRNA array showed that, compared with normal thyroid, there had been 39 and 25 miRNAs different expression in HT and HT side of HT association with papillary thyroid carcinoma(PTC), respectively(P < 0.05); miR-142-3p, miR-338-3p, miR-454, miR-146 a,miR-29b-1*, miR-150 and miR-223 got the same miRNAs up-regulation(log2 FC≥2,P<0.05). while miR-654-5p, miR-601, miR-198 and miR-1226*(log2 FC≥2, P<0.05)got down-regulation. In addition,miR-142-5p increased nearly eight times higher in primary HT than the normal thyroid(log2 FC =7.959, P <0.01). Conclusions: With the method of micro RNA microarray, we found the related miRNA expression profiles of the Hashimoto’s thyroiditis, general initially mastered and compared the miRNA expression profiles of Hashimoto’s thyroiditis with normal thyroid, which laid a foundation for the further functional research of the disease-associated miRNAs.
