Abstract: 目的 优化与改进匹伐他汀钙中间体的工艺条件.方法 采用3-环丙基-3-氧代丙酸乙酯(1)与2-氨基-4'-氟二苯甲酮(2)缩合得到2-环丙基-4-(4'-氟苯基)-3-喹啉甲酸乙酯(3),将酯(3)水解为2-环丙基-4-(4'-氟苯基)-3-喹啉甲酸(4),再将(4)还原为2-环丙基4(4'-氟苯基)-3-喹啉甲醇制得匹伐他汀钙重要中间体母核醇(5).结果 工艺改进后,嘧啶甲基醇衍生物5的总收率由55.3%提高到77.1%.结论 新工艺通过对反应条件的优化,使母核醇5的合成操作简单,成本低,更适合工业化生产.
