EXPRESSION PATTERN OF COMPLEMENT-RELATED GENES IN NORMAL AND LPS-CHALLENGED ZEBRAFISH EMBRYO/LARVA

Abstract: To understand their roles in early development and maturation of complement system in zebrafish, we studied the expression patterns of several complement genes(C3, C4, C9, Bf1, Bf2, Bf3, RCA2.1, and RCA2.2) and their responses to LPS-induced challenges, using WISH and q RT-PCR techniques. The result demonstrates that both complement and complement regulatory genes were contributed maternally, expressed ubiquitously during early developmental stages(from cleavage stage to segmentation stage), and then expressed mainly in the liver and digestive tract in later developmental stages. C3, C9, Bf1, Bf2, and Bf3 genes were all up-regulated in embryos/larvae after LPS injection, while RCA2.1 gene was down-regulated. Therefore, zebrafish embryos are capable of activating and regulating complement system to protect them from pathogenic infection.
