Qiang Gu Kang Wei Prescription Increases Bone Mineral Density of Load-bearing Bone in Tail Suspended Rats

Abstract: Objective To investigate the effects of traditional Chinese compound prescription-Qiang Gu Kang Wei Prescription( QGKWP) on bone metabolism of tail suspended rats. Methods Twenty one male SD rats were divided into control group( Con),the tail suspension group( TS) and QGKWP treatment group( QGKWP) randomly. Rats were treated with 2. 5 mL saline or 30 mg / g QGKWP by gavage every morning and afternoon,respectively. The bone mineral density( BMD) of rats was detected with dual energy X-ray absorptiometry every week. After 28 d of test,blood serum was collected for determination of alkaline phosphatase( ALP),bone alkaline phosphatase( BALP),bone gamma-carboxyglutamic acid-containing protein( BGLAP)and deoxypyridinoline( DPD). The mechanical property of weight-bearing bone,bone dry weight and bone ash weight were observed. Collagen I,osteocalcin and osteopontin were detected with immuno-histo-chemistry( IHC) staining. Results Compared with TS,QGKWP significantly increased the BMD of femur,tibia and lumbar vertebrae. QGKWP also promoted serum ALP and BGLAP level. Quantitative IHC analysis showed that QGKWP increased the expression of collagen I and osteopontin in femur of TS rats. Conclusion QGKWP partially reverses the effects induced by TS and may be a potential countermeasure for bone loss.
