Extraction Technique of Protein from Proso Millet

Abstract: Effects of main factors, including extraction time, extraction temperature, ratio of material to liquid and p H value on proso millet protein yield were investigated. Process conditions of extracting protein from proso millet by alkali-solution and acid-isolation were optimized using orthogonal experiments. Results showed that extraction factors had effects on protein yield.Effect order of independent factor on protein yield was p H>ratio of material to liquid > extraction time > extraction temperature. p H value was the most important factor influencing protein yield, followed by ratio of material to liquid and extraction time. A maximum protein yield of 86.93% was achieved at extraction time 80 min, extraction temperature 30℃, ratio of material to liquid1/2.5 and p H 10.
