Abstract: 采用苯丙氨酸解氨酶(PAL)为模型酶,制备了以粗孔微球硅胶为内核的苯丙氨酸解氨酶交联酶聚体(NH-PAL-CLEAs),优化了制备条件.并研究了NH-PAL-CLEAs的部分催化性能.研究结果表明,当硅胶量为117.75 mg,酶液添加量为1 mL(2 U),戊二醛浓度为0.2%(体积分数)时,所制备的NH-PAL-CLEAs的最大酶活回收率是25%.与传统的PAL-CLEAs相比,NH-PAL-CLEAs无需离心或过滤,只用短时间静置就能被回收重复利用.同时,NH-PAL-CLEAs也表现出较好的操作稳定性.这种制备方法不仅克服传统制备交联酶聚体(CLEAs)颗粒小、难回收,离心或过滤回收易结块的问题,而且缩短了生产周期,减少了能源消耗,提高了固定化酶的利用率,是一种非常具有工业应用前景的固定化酶方法.
