Effects of acitretin on the synthesis of nitric oxide and the proliferation of HaCaT cells induced by sub-stance P

Abstract: Objective:To determine the effects of acitretin on the synthesis of nitric oxide ( NO) and the proliferation of HaCaT cells induced by substance P ( SP ) . Methods:The cells were classified into the blank group, SP group, low dose acitretin combined SP group, moderate dose acitretin combined SP group and lar-ger dose acitretin combined SP group. The level of NO in the supernatant of cell culture medium of HaCaT cells were detected by nitric aciddeoxidize enzyme method after culture for 30 minites. The proliferation rate of HaCaT cells in all groups was detected by MTT. Results:Compared with SP group, the level of NO in the low dose acitretin combined SP group was higher and that in the high dose acitretin combined SP group was lower ( P<0.05) . The proliferation of HaCaT cells in the low dose acitretin combined SP group and moderate dose acitretin combined SP group was higher and in in the high dose acitretin combined SP group was lower than that in the SP group ( P<0.05) . Conclusion:Acitretin may had the bidirectional regulation on the synthesis of NO and the proliferation of HaCaT cells.
