The application of DNA barcoding identification kit technology to Monochamus Dejean

Abstract: In order to rapidly and accurately identify the Monochamus of wood from Russian, we employed DNA barcode detection kit technology and designed a pair of nested primers based on mitochondrial COⅠ gene ( mitochondrial cyto?chrome oxidase subunit I) . Furthermore, COⅠ gene sequence of eight Monocharnus species was amplificated, and up?loaded to pest information site. According to differences of arrangement and composition among these species at base sites, species unique bases was got in the corresponding sites, which is named specific site to distinguish this species. In this way, we developed the DNA barcoding identification kit technology to Monochamus species, which will provide a new way for port quarantine identification.
