Abstract: Objective To investigate the effectand mechanismof hypoxiinducible factor-1α (HIF-1 α) on lipid metabolism in prostate cancecells.MethodThe expression of HIF-lα and lipogenigenewere compared in differenprostate cancecell lineby real-time fluorescenquantitative polymerase chain reaction (FQ-PCR) and Western blotting.Then the expression of lipogenigenein prostate cancecellundehypoxienvironmenotransfected with HIF-1α-Mutanplasmid waanalyzed.In addition, luciferase assay and ChIP assay were used to determined the underlying moleculamechanisms.ResultThere isome correlation between HIF-1α and lipogenigenes.Hypoxienvironmenand overexpression of HIF-1α could increase the level of lipogenigenes.HIF-1α could bind to the promoteof sterol-regulatory elemenbinding protein(SREBP)-1and regulate ittranscription.Compared to negative control (1.000 ±0.095, 1.000 ±0.042), HIF-1α could significanincrease the mRN(2.992 ±0.159) and protein (2.349 ± 0.114) levelof SREBP-1 c.Conclusion In hypoxienvironment, HIF-1 α could promote de novo ligogenesithrough promote the transcription of SREBP-1in prostate cacnecells.
