Abstract: 为了获得恶性疟原虫红细胞内期不同生长阶段更加精准的组织材料,本研究探索一种简便易行并能获得同步化程度很高的恶性疟原虫虫株的方法.该方法采用山梨醇多次连续同步化恶性疟原虫环状体,并继续培养得到同步化程度高、窗口小、虫血率高的红细胞内期各期的恶性疟原虫虫株;室温条件下(17~20C)继续培养同步化至裂成熟殖体期的恶性疟原虫虫株,用孔径1.2 μm的无菌滤膜过滤,收集裂殖子.经同步化处理之后,恶性疟原虫在环期、早期滋养体期、晚期滋养体期和成熟裂殖体期的同步化程度分别在90%、85%、85%和80%以上,同步化后各期恶性疟原虫虫血率可达10%~20%,各期虫株的生长周期的窗口可达4h以内.
