Rapid Screening and Breeding of High Yield Strains of Cordyceps militaris by Using Double-Layer Agar Plates

Abstract: According to the prevalent strain degeneration phenomenon in the actual production,the double-layer agar plates(DLAPs) method for rapid screening and breeding of high yield strains of Cordyceps militaris are studied. The selective medium(code C),ordinary medium(code N) and enriched culture medium(code R) are pairwise combined. DLAPs are used to screen and breed degradation bacteria mixed conidia,and compared with the traditional plate streaking method. Bases on the comparison results of mycelium germination,colony morphology,mycelia density,enlarge cultivation and production inspection,the CR plate(the bottom layer is selective medium,and the upper layer is enriched culture medium) is selected.The CR plate shows good isolation effects of the strains,and provids high proportion of high quality pure strains with good characters,high activities,strong fruiting body and high bud differentiation rates. It can effectively reduce the workload of screening and provide a guarantee for the industrial production of stable strains.
