Abstract: Objective To investigate effects of subcutaneous injection of isoprenaline ( ISO) on cardiac hypertrophy in mice.Meth-ods Male C57 mice were divided into two groups:normal saline group ( NS group) and ISO group.The mice were subcutaneously injec-ted with a dose of 5mg/kg of ISO 2 times a day for two weeks, meanwhile mice in the NS group received the same volume of vehicle.The ejection fractions(EF%)were recorded after mice cardiac ultrasound.The heart weight(HW), lung weight(LW), and body weight(BW) were recorded and then the ratios of HW/BW and LW/BW were calculated.The cross-sectional areas ( CSA) of the myocytes was also counted.Atrial natriuretic peptide ( ANP) was detected by real time PCR.The signaling pathway involved in the cardiac hypertrophy was also detected by western blot.Results ISO induced reduced EF%,induced increased HW/BW, LW/BW, CSA of myocyte and mRNA level of ANP.And expressions of phospho-ERK and phospho-JNK in ISO group were enhanced compared with NS group.Conclusion Subcutaneous injections of ISO can effectively induce cardiac hypertrophy in mice model through the activation of MAPK signaling pathway.
