Abstract: Unasyn is in combination of ampicillin with sulbactam. The serum bactericidal activity (SBA) was studied by crossover control design in ten health volunteers receiving ampicillin 2g or unasyn 3g (containing ampicillin 2g and sulbactam 1g) given iv. The serum samples collected 30 min and 6 hr after administrati on were measured by a microtiter system using Mueller-Hinton broth as the diluent.SBAs of unasyn against the strains of S.aereus, Pr.mirabilis, Acinetobacier catcoa-ceticus and K. pneumonia producing β-lactamase and resistant to ampicillin were significantly higher (l:8->l:32) than ampicillin alone(<l:l-l:2). %SBA>1:8 of unasyn against mentioned above strains were 50~100% and displayed very good bactericidal activity. SBAs of unasyn against ampiciilin-resistant strains of E. coli and E. cloacae were slightly low, but still higher than ampicillin. Both ampicillin and unasyn showed rather weak bactericidal activity against Ps, aeruginosa.It is suggested that combination of sulbactam with ampicillin showed synergetic activity against offending pathogens producing β-lactamase and resistant to ampicillin butPs. aeruginosa.
