Abstract: The release of Penicillin acylase from the intra of E, coli cells by means of treatment of organic solvent and refrigeration at - 30℃ was studied and the enzyme releasing ratio can be up to 77%. The method of NIPAB analyzing the Penicillin acylase is improved.The purification of Penicillin acylase by using two-aqueous system PEG 4000 and Potassium phophate is developed. The optimum constitution of the system is determined, which is composed of 16% (w/w) PEG 4000 and 14% (w/w) Potassium phosphate. The activity recovery ratio of this intracellular enzyme from the released solution is more than 90% and the purification factor can be up to 311.
