Abstract: The present study deals with a novel anticancer antibiotic,actinomycin 23-21,isolated from Fuzhou soil.HeLa cells were cultured in the medium consisting of RPM1 1640 and bovine serum,and treated with different concentrations of the drug. According to the dose-response curve,the concentration corresponding to 50% survival rate was taken for experimental use,which contained actinomycin 23-21 20μg/ml. The growth curve of HeLa cells treated with the drug and their growing ability on soft agar all indicated that actinomycin 23-21 could inhibit the growth of HeLa cells. Reproduction and colony formation of HeLa cells were also inhibited by the drug. Some structural changes such as swelling and breaking-up of HeLa cells treated with the drug were observed by microscopy. Clumping of peripheral heterochromatin,shrinkage of nucleolus,and separation of nucleolus with nucleo-associated chromatin could be seen under Electron microscope(×10,000). Immunosuppressed mice inoculated with HeLa cells treated with actinomycin 23-21 did not develop neoplasms. Meanwhile,epithelioma developed in the mice of control group. All of these results confirmed the inhibitory effect of actinomyciu 23-21 on HeLa cell line.
