Abstract: This paper dealt with the gas chromatographic analysis of N-cyclop-entyl-N’-amino-piperazine——the side chain of long-acting antituberculosis antibioticRifapentin.Experimental results showed that the side chain and other 13 related piperazine compounds in the sample could be completely resolved by using 2% OV-17 on chromosorb W (60-80 mesh)as stationary phase.A derivatization method was proposed due to the labile property of the side chain, by condensing the amino group of the side chain with benzaldehyde to form Ncyclopentyl-N’-benzalimide-piperazine.This method is accurate, rapid and simple. It can be used for quantitative analysis of the side chain and is superior to silyation and acetylation method. The derivatization procedure reacted quantitatively, and the product is a stable solid compound which is easily purified.The quantitative standard deviation is 0.6 and relative standard deviation is 0.7%.
