Abstract: The penicillin acylase was extracted from Escherichia coli by method of solvent-freeze thawing, and purified by precipitation with （NH4）2SO4, then dialysed and freeze-dried. The extraction yield was 47. 93%.The Kinetics Michealis-Menten’s constant Km for penicillin acylase and its immobilized enzyme were estimated.The measurements were made at the influence of various values of pH, temperature and metal ions.
