Optimization of process parameters for straw cellulose ethanol production based on response surface methodology

Abstract: This paper optimized the culture conditions for cellulase production by Trichoderma reesei, which based on the single factor optimization results, with filter paper activity(FPA) as response values, and three factors that had the most significant effect on enzyme production were screened through the Plackett-Burman analysis,which as follows: wheat bran > temperature > calcium chloride. The results of response surface methodology optimization showed that the theoretical maximum cellulase activity was 62 281.3 U/mL when wheat bran concentration, temperature and calcium chloride concentration were 6.27 g/L, 31 ℃ and 0.709 g/L,respectively. Under the optimized conditions, the practical FPA of crude cellulase was 60 126.5±16.0 U/mL. Then crude cellulase with10% adding quantity was added to 5 L fermentation tank of converting straw into ethanol, the yield of ethanol(v/v)reached 7.05%±0.18% after 144 h.
