EFFECT OF MAYTENUS HOOKERⅡ LOAS ON THE FINE STRUCTURES IN THE CELLS OF AN ESOPHAGEAL CARCINOMA CELL LINE

Abstract: Maytenus hookerii Loas is a new experimental antitumor drug.Its effect on the fine structures in the cells of the ECa 109 esophageal carcinoma cell line herein is described.After treatment with the drug,some of the cells show vacuolar degeneration and others become multinucleated with many micronuclei.The“confronting cisterna” of the endoplasmic reticulum is conspicuous in many cells.Polyribosomes,lysosomes, multivesicular bodies and nuclear pores are frequently visible.Golgi bodies become dispersed throughout cytoplasm.Glycogen particles can be seen in some and kinetochores and centrioles are found in many of the cells.Only a few spindle tubules are observed but bundles of fine filaments are frequently seen.The above features resemble the c-mitosis effect produced by colchicine.These observations indicate that Maytenus blocks cell division by disturbing the spindle tubule system,eventually leading to abnormal chromosome movement.The ultrastructural studies on these cells suggest that Maytenus hookerii Loas may interfere with the structure and function of microtubules,both in interphase and in mitosis and can be regarded as a rather specific microtubular inhib- itor. As Maytenus hookerii Loas acts mainly upon mitotic cells,it may be used as a cell cycle specific drug in the combination chemotherapy of cancer.
