Antinociceptive effect of bergenin on vincristine-induced neuropathic pain?

Abstract: Obj ective :To investigate the antinociceptive effect of bergenin on vincristine-induced neuropathic pain (CNP)and to explore its mechanisms. Methods : Mechanical withdrawal threshold was tested before and after intraperitoneal administration of 100 mg/kg bergenin in the model mice with vincristine-induced neuropathic pain via behavioral experiment. Expression of oxidative stress markers including malondialdehyde and glutathione was determined biochemically.Ca2 + level was detected by atomic absorption spectrometry.Sciatic nerve production of TNF-αand IL-1βwas quantified by ELISA kits. Results :Bergenin significantly attenuated vincristine-induced mechanical allodynia.Bergenin also ameliorated vincristine-induced high oxidative stress,inhibited the expression of TNF-α and IL-1β,and reduced the Ca2 + level in CNP mice. Conclusion:Bergenin may attenuate vincristine-induced neuropathic pain.The effect might partly attribute to the inhibition of oxidative stress and the decrease in the inflammatory cytokines expression as well as Ca2 + level.
