Study on HPLC Fingerprint of Amino Acids Compositions of Panacis Quinquefolii Radix and Dendrobii Caulis in Medicated Diet

Abstract: Objective:To establish the HPLC fingerprint of amino acida compounds from Panacis Quinquefolii Radix and Dendrobii Caulis in medicated diet.Methods:To apply 2,4-initrofluorobenzene pre-column derivatization HPLC method.Separation was perfomed on Kromasil C18(250mm×4.6mm,5μm)analytical column with mobile phase consisting of N,N-dimethylformamide-0.025mol·L-1 sodium acetate(1∶100,with 36%acetic acid adjust to pH6.0)and acetonitrile with the flow rate of 1.0mL·min-1 and the column temperature at 40℃.The UV detection wave length was set at 360nm,the injection volume was 10μL and 10batches were determined.Results:The fingerprint of Panacis Quinquefolii Radix and Dendrobii Caulis in medicated diet was established with 23common peaks,17kinds of amino acid compsdition were identified,The similarity of 10batches of samples was above 0.92. Conclusion:Each ingredient’s amino acids showed in the characteristic fingerprint.The characteristic fingerprint of amino acid components can be used for the quality control.
