Optimization of Ultrasonic Extraction Technology of Flavonoid Glycosides in Scutellaria Baicalensis by the Combination of CCD-RSM and a Two-indicator Model

Abstract: Objective:To optimize ultrasonic extraction Technology of baicalin and wogonoside in Scutellaria baicalensis by central composite design-reponse surface method(CCD-RSM).Methods:The contents of baicalin and wogonoside were determined by Ultra performance liquid chromatography(UPLC).Independent variables were ethanol concentration,liquid-solid ratio and extraction time,the dependent variable was the overall desirability of extraction rate of baicalin and wogonoside.Linear or no-linear mathematic models were established to estimate the relationship between independent and dependent variables.Response surface methodology was used to optimize the process of extraction.Prediction was carried out through comparing the observed and predicted values.Results:The optimum extraction technology was as following:The extraction time was 45 min with solvent to solid ratios(v/w)of 40:1,and the extraction solvent was 58% ethanol at the extraction temperature of 40℃.The deviation between the observed and predicted values was small(P<0.01).Conclusion:This optimized extraction technology was stable and feasible with high predictability.This method can be provided reference for exploring the flavonoid glycosides in Scutellaria baicalensis.
