Abstract: To provide technical references for protection of wild E.acuminatum resources and in vitro production system of flavonoids,the young leaves of E.acuminatum were used as explants for callus induction and its factors including different sterilization agent, medium, hormone combinations and illumination conditions which affect the callus induction of E.acuminatum were studied.Besides,the content of flavonoids in the calli was studied by means of the ultraviolet spectrophotometric analysis.The results indicate that the callus culture of E.acuminatum leaves can be induced under the following conditions:disinfection of 2% NaClO for 14 minutes,the medium MS + 2,4-D 2.0 mg/L+ 6-BA 0.5 mg/L+ IBA 0.5 mg/L + NAA 1 mg/L,at 24℃ 1 000 lx illumination conditions.The subculture and preservation is much appropriate under dark conditions.Total flavonoid content of induced embryonic calli is up to 5.38%,higher than that of pharmacopoeia standard.
