Abstract: 目的研究吡那地尔对癫痫持续状态(SE)大鼠海马神经元保护作用及可能机制。方法随机将96只大鼠分成N组、SE组、Pin组、Gli组,每组按SE后12h、24h、72h、5d分为4亚组,用LiCl-PILO法进行SE动物造模,采用SABC检测Bcl-2、Caspase-3,TUNEL法检测凋亡细胞。结果N组TUNEL阳性细胞、Bcl-2及Caspase-3均有基础表达。SE组、Pin组、Gli组TUNEL阳性细胞均在72h达峰值、Caspase-3均在24h达峰值。同时间点比较,SE组、Pin组、Gli组较N组均明显增多(P<0.05);Pin组较SE组、Gli组均明显减少(P<0.05);SE组与Gli组比较差异无统计学意义(P>0.05)。SE组、Pin组、Gli组Bcl-2均在12h达高峰。同时间点比较,SE组、Pin组、Gli组较N组均明显增多(P<0.05);Pin组较SE、Gli组均明显减低(P<0.05);SE组与Gli组比较差异无统计学意义(P>0.05)。结论吡那地尔可上调Bcl-2表达,抑制Caspase-3释放,对SE大鼠有脑保护作用,格列吡嗪拮抗吡那地尔此作用。
