Abstract: Separation of Mexiletine hydrochloride enantiomers by HPLC was established on C18 column using the carboxymethyl-β-cyclodextrin ( CM-β-CD in short) as chiral mobile phase additive. The influences of concentration of chiral mobile phase additive, the organic modifier, the pH value and the flow rate on the enantiomers separation were investigated systematically. The better separation was achieved when the mobile phase consist of methanol-acetoni-trile-water 15∶70∶15(V/V/V), pH=4. 28( adjusted by triethylamine-glacial acetic acid ), containing 0. 5 g· L-1 CM-β-CD, at the flow rate of 0. 3 mL·min-1 . The detection wavelength was set at 212 nm.
