Probing into Effective Mechanism of Yishen Zhishuai Granules on Renal Function of 5 /6 Nephrectomy CRF Rats by Utilizing the Method of “Centralizer and Descending Turbidity”

Abstract: Objective: To investigate the effective mechanism of Yishen Zhishuai Granules improving renal function of rats with renal failure. Methods: Selecting Sprague-Dawley rats,we made rats models of chronic renal failure by 5 /6 nephrectomy.After two weeks of the operation,we drew the blood from inner canthus,and tested model group according to the serum creatinine level. The rats models were randomly divided into: B as model controlled group; C as Yishen Zhishuai Granules group; D as positive controlled group( Losartan Potassium Tablets); no operation rats were A as false operation group. After 3days of grouping,A and B group will be given distilled water( 3ml / d); C,D and E group will be given the following corresponding drugs( 10 times the adult standard dose) by lavaging continuously; C: deploying solution( containing raw materia of Yishen Zhishuai Granules 3. 2g / ml); D: Losartan Potassium( Cozaar) 8. 6mg / kg / d; After 56 days at giving drugs,we would kill rats and test urea nitrogen,creatinine by kit; expression level of TGF-β1,HGF,BMP-7,WT1 mRNA of residual renal tissue by real-time PCR method. And observed the lesions of residual renal tissue under light microscope after hematoxylin-eosin staining.Results: By intervening rats models of chronic renal failure by 5 /6 nephrectomy,we preliminarily found positive control drug,Yishen Zhishuai Granules all could improve renal function in varying degrees. With testing residual renal tissue,by real- time PCR method,Yishen Zhishuai Granules could down-regulate expression of TGF-β1mRNA obviously,and up-regulate expression of HGF,BMP-7mRNA obviously. Compared with the model controlled group,obvious expression with HGF mRNA but no statistic significance with low expression of TGF-β1mRNA and BMP-7mRNA in Losartan Potassium group. There was statistic significance between Yishen Zhishuai Granules and Losartan Potassium group. Both could up-regulate positive expression level of WT1 mRNA obviously. The each of treatment groups reduced pathological damage of residual renal tissue in different degree under light microscope after hematoxylin-eosin staining. Conclusion: Yishen Zhishuai Granules can improve renal function of rats with renal failure by 5 /6 nephrectomy,the effective function is relation to down-regulating expression of TGF-β1mRNA,up-regulating expression of HGF mRNA,BMP-7mRNA,which can increase expression of WT1 mRNA in the tissue are increased. Preliminary studies have showed Yishen Zhishuai Granules can impair balance disorders between TGF-β1and HGF,balance disorders between TGF-β1and BMP-7; and regulate balance of renal tissue remodeling and fibrosis,so that improve renal function of rats with CRF by 5 /6 nephrectomy,which is one of mechanism that Yishenzhishuai granules can delay the course of CRF.
