Abstract:
以两个普通小麦耐盐突变体8901-17和974915及其各自的亲本为材料,依据前人的工作选择第一、三、四、六同源群染色体上的生化标记进行了耐盐突变体突变位点的定位研究.高分子量麦谷蛋白亚基(HMW-GS)SDS-PAGE,EST-5、β-Amy-1、α-Amy-1等电聚焦(IEF)电泳分析结果显示:974915与其父母本在HMW-GS、βAmy-1谱带上表现出了明显的差异带型;8901-17与其亲本冀麦24在α-Amy-1谱带上表现两条与耐盐性有密切关系的差异带.两个突变体与其相应亲本在EST-5上均未表现出差异.由此可见,突变体974915的突变位点主要发生在第一、四同源群上,而8901-17的突变位点则主要发生在第六同源群上.该结果为RFLP定位耐盐基因时选择合适的探针提供了依据.
